昆虫免疫防御系统的限制因素 Ξ

摘要 :本文主要针对昆虫对寄生物的免疫防御并从进化生态学的角度来阐述昆虫防御系统所存在的一些限制因素 ,如识别
抗原失败、免疫特异性、防御代价、自体免疫、性别、资源利用率和寄生物的干扰等 ,而这些局限使得昆虫的免疫防御受到不同 (自
发或诱导的、广泛或具体的)免疫反应的影响 ,从而限制其持续的免疫防御。

Limitations of the Insect Immune Defense

ABSTRACT : In this review , some limitations of insect immune defenses especially with respect to parasitism were introduced from the evo2
lution ecology’s aspect , including the failure to recognize an antigen , response specificity , the cost of defense , and the risk of autoimmunity ,
sex , resource availability , and interference by parasites. Such limitations affect the insect immune defenses in different aspects (unmediated or in2
duced , general or specific) , and in turn restrict the persistent immune defenses of insect.

1 引言
研究发现昆虫免疫防御存在一些限制因素 ,如识别抗原
失败、免疫特异性、防御代价、自体免疫、性别、资源利用率和
寄生物之间的干扰。而这些局限使得昆虫的免疫防御由不同
(自发或诱导的、广泛的或具体的) 的免疫反应构成 ,从而影响
到其持续的防御。这种情况对于昆虫来说究竟是利大于弊还
是弊大于利需要数学方法进行分析 ,然而这种研究在目前还
少有进行。总的说来 ,昆虫免疫防御与脊椎动物的免疫防御
极其相似 ,而且这个研究领域正在随着分子生物学研究技术
的发展而日新月异。
2 昆虫的免疫防御
2. 1 昆虫的免疫防御系统
昆虫的免疫防御是指昆虫排斥外源抗原异物的能力 ,体
现在其自身的免疫防御反应。主要由昆虫的血淋巴系统承
担 ,分为细胞免疫和体液免疫。其中血细胞主要承担细胞免
疫 ,起着吞噬作用、成瘤作用、包囊作用和凝集作用。血浆则
承担体液免疫 ,主要分为先天性免疫 (起作用的主要是血浆中
的一些溶菌酶和酚氧化酶)和后天性免疫 (昆虫在被感染之后
所产生的抗菌肽) 。图一是昆虫识别抗原后进行免疫反应的
一般

2. 2 昆虫免疫防御系统的特点
研究
明昆虫免疫系统中的一种关键物质在哺乳动物免
疫系统中同样存在 ,这
两类生物的免疫系统是同宗的 ,很
可能来自于 5 亿多年前的共同祖先 [1 ] ;美国科学家的研究还
表明昆虫的免疫防御系统是一个复杂的系统 ,远远超乎学者
以往的想象[2 ] 。另外更加重要的是昆虫免疫防御系统并不能
产生真正的抗体 ,当虫体受到某种寄生物侵染时 ,其机体内并
不能产生真正的抗体而进行抗体 - 抗原免疫反应 ,正因为如
此也导致昆虫的免疫防御系统不具有记忆效应 [3 ] 。然而 ,昆
虫在漫长的演化过程中也形成了它自身独特的免疫机制 ,其
显著特征是一旦被外源抗原异物侵入时 ,体内迅速地产生抗
菌蛋白 (如抗真菌肽和抗细菌肽等) ,但是这种蛋白并不象高
等动物体内产生的抗体那样能够永久存在 ,只能存在一定的
时间不久便会消失 ,而当昆虫再次遭到同种外源抗原异物的
感染时 ,体内又会产生同样的抗菌蛋白 ,因此昆虫免疫防御的
持效性比较差[3 ] 。
3 昆虫免疫防御的限制因素
3. 1 资源利用率
目前这方面的研究较少且几乎集中在昆虫的食物与其免
疫防御间相互关系的研究上。例如 ,长红猎蝽 ( Rhodnius pro2
lixus)的幼虫如果仅以血浆为食就会比以完整血液为食的幼虫
产生更少的抗菌肽、溶菌酶活性降低、血细胞数目更少并且在
实验室条件下对于细菌的感染抗性更低 [4 ] 。也有实验表明在
一定的饲养条件下 ,抗菌肽的产生量由昆虫体内的脂肪储量
决定[5 ] ,脂肪的储量与昆虫所摄取的食物密不可分。长期的

食物匮乏会导致黄粉虫 ( Tenebrio molitor) 免疫功能规律性下
降[6 ] ,也能导致舌蝇对锥体虫感染的敏感性上升 [7 ] 。然而 ,严
重的食物匮乏并不影响熊蜂包囊异物的能力 ,尽管由于食物
的匮乏急剧地降低了它们生存的适合度 [8 ] 。
3. 2 免疫特异性和寄生物的干扰
寄生物在侵入寄主体内后能抑制寄主的免疫防御 [9 ] ,在
本文中这种抑制称为寄生物的干扰。比如 ,拟寄生蜂会使用
病毒作为一种“生物武器”来抑制寄主的免疫 [10 ] 。还有报道证
实锥体虫在侵染其介体寄主 (Rhodnius) [11 ]以及线虫 (Steinerma)
在侵染蜡螟 ( Galleria) [12 ]的时候都会抑制寄主的酚氧化酶。寄
生物在抑制寄主的免疫防御时大多直接向寄主体内注射毒
素[13 ] ,然而抑制方式随拟寄生物的基因型、地理种群以及寄主
种类的不同而不同 ,这种现象说明寄生物与寄主之间存在着
一种协同进化的关系 [14 ] 。
昆虫免疫防御的识别
也会影响免疫防御反应。如果
昆虫免疫系统的识别分子只能很笼统地识别某些种类的寄生
物 ,如革兰氏阴性细菌和革兰氏阳性细菌 ,那么有大量的抗原
就将会被昆虫识别而不加选择地排斥。但是通过侵染试验发
现寄主昆虫的免疫防御也具有很强的专化性 ,昆虫能辨别不
同的寄生物甚至是同一寄生物的变异体 (包括物理的变
形) [15 ] ,这种免疫的特异性就使得许多抗原对昆虫的免疫防御
系统失去了刺激作用 ,因而不能激活免疫反应从而限制昆虫
的免疫防御。
另外在昆虫体内还发现了交互效应 ,昆虫对某一种寄生
物产生免疫反应就会诱导其自身免疫系统抵御其它种类的寄
生物[16 ] ,尽管这种诱导的免疫防御方式既不彻底又不相同于
诱导其产生的免疫防御方式 [17 ] 。这种交互效应能带给昆虫较
为长期的保护作用 ,这说明在昆虫被第一次感染以后其免疫
系统的敏感性会增加 [18 ] 。有实验证明交互效应的保护作用产
生的途径可能与许多抗原引起昆虫免疫防御的途径是一样
的 ,因为实验观察到昆虫的抗菌肽对许多的微生物都具有生
物活性[19 ,20 ] 。尽管在这些试验中采用的都是比较粗略的方法
来研究这样的免疫反应 ,但至少也说明一点 ,即这种防御途径
的变化必然是某个特定的免疫防御系统被激活后发生的。显
然 ,目前昆虫专化免疫反应的研究是一个比较广阔的领域 ,因
为到目前为止 ,昆虫乃至动物中基于共同的异物识别系统、共
同的中间途径或者共同的分子防御机理的交互免疫性还知之
甚少。
3. 3 防御代价
进化生态学研究发现昆虫免疫防御的代价是限制昆虫免
疫防御的一个重要的因素。昆虫的免疫防御不仅要消耗能量
和食物 ,而且还会造成昆虫在进化过程中其体内某些与生存
适合度有关的功能下降。另外 ,有学者认为昆虫维持其免疫
防御能力需要代价 ,并且这种代价在维持昆虫的繁殖能力与
其存活率的平衡上是必不可少的 ,这种平衡是昆虫生活史理
论的一个中心原则 [21 ,22 ] 。因此 ,本文将从以下两方面 (进化代
价和维持防御的代价)简述昆虫免疫防御代价。
3. 3. 1 进化代价。当昆虫自身选择了一个高强度的免疫防
御系统传递给其后代时 ,由于基因的多效性和遗传协方差 ,昆
虫体内一些与适合度有关的功能相应地就会减弱。比如 ,寄
主昆虫的一些品系不易被寄生物感染时通常生殖能力下降 [23 ]
或者是其后代发育历期变长 [24 ] 。还有研究表明将一些寄主昆
虫的品系用来进行抗寄生物能力研究时 ,这些昆虫其它的一
些功能就大大地被削弱了。如蜜蜂在遭遇幼虫腐败病 (蜜蜂
的一种毁灭性疾病) 后 ,其后代幼虫的发育速率就降低 [25 ] ;对
颗粒体病毒产生抗性的印度大斑粉螟后代发育历期延长并且
卵的孵化率下降 [26 ] ;对普通寄生物产生抗性的果蝇在食物的
争夺中其竞争力下降。反之 ,其它功能若得以强化其免疫防
御能力就下降。如生殖能力强的黄粪蝇 (Scatophaga stercoraria)
其后代体内酚氧化酶活性就降低。
因此 ,实验表明强大的免疫防御系统在进化的时候可能
是伴随着其它与适合度相关功能的减弱而进行的 ;同时 ,其它
功能的进化也可以导致免疫功能的下降 [27 ] 。虽然这些代价的
本质还不清楚 ,但是有学者认为这有可能是昆虫体内抗寄生
性的某个受体 (如果蝇中的 Toll 蛋白) 与其生长发育有着密不
可分的联系[28 ] 。
3. 3. 2 维持防御的代价。昆虫的防御能力要能得以维持就
必须付出某种代价 ,这种代价与前面进化代价有些相似 ,因为
从生理学上来说 ,生物要维持其强大的免疫能力就必定要弱
化其它功能。然而 ,维持防御的代价却是不稳定的。昆虫表
皮黑化程度呈季节性变化就说明了这一点 ,这昆虫表皮黑化
程度的变化可以视为反映了昆虫要么是在冬天开始时其免疫
防御能力上调的一种表现 ,要么就是经历了之前生殖季节后
其生殖能力下降的一种表现 [29 ] 。当昆虫的免疫反应被激活以
后其耐旱性和耐饥性都下降[30 ,31 ] ,将脂多糖 (LPS) 注入熊蜂
(Bombus terrestris)体内时 ,其整个种群的存活率下降 [32 ]而且种
群生存的适合度也降低 [33 ] ;注入脂多糖 (LPS)也能降低冈比亚
按蚊 (Anopheles gambiae)的繁殖能力[34 ] ,同样的处理方法也能
增强菜粉蝶蛹的代谢速率 [35 ] 。反之 ,当昆虫进行其它能量消
耗大的活动时 ,其免疫防御能力就下降 ,比如豆娘包囊作用减
弱[36 ] 、以及雄性蟋蟀体内酚氧化酶活性和抗细菌感染力下降
时 ,它们的繁殖能力就显著增强 [37 ,38 ] ;被迫地搜寻食物会使得
熊蜂工蜂包囊体内所注入异物的能力下降 [39 ] 。由此可见 ,昆
虫的免疫防御能力要得以维持就必然付出相应的代价。
3. 4 自体免疫
昆虫的免疫防御要依赖于对自身物质和异己物质的正确
识别 ,正因如此才容易引起识别失败的问
。如果识别的标
准太过单一苛刻就容易使大量的病原物逃脱 ;相反如果识别
的标准太过宽泛就不仅能识别病原物 ,而且连自身物质都难
逃厄运。无脊椎动物的自体免疫问题已经在多篇文章中提
到[27 ,35 ,40 ] ,但是现在对于昆虫自体免疫机理和自体免疫影响
的研究仍然很缺乏。这方面比较初级的信息来自果蝇的研
究 ,当血细胞聚集成囊状并对自身组织产生免疫反应时果蝇
体内就会形成黑化的瘤 [41 ,42 ] 。在果蝇中这种潜在的组织坏死
和黑化可能与丝氨酸蛋白酶抑制蛋白和先天免疫的调节蛋白
有关[43 ] 。
昆虫免疫系统具有一些与生俱来的特点 ,这些特点可以
导致自我损害。例如昆虫的循环系统是一个开放性的系统可
以运输免疫系统产生的分子使得昆虫的免疫防御系统似乎更
为有效 ,但正因如此也使得其身体的每个部分都有可能受到
潜在的危害。因为免疫分子通常有比较多的靶标 ,比如免疫
过程中产生的醌就能与氧和氮结合从而造成普遍的细胞损
伤[44 ] 。
当然昆虫也具有许多的调节机制可以减少这种自动防御

反应带来的损伤。其中一个很重要的机制就是出现在昆虫肠
道内的将自动防御反应停滞的这个机制 [45 ] ,这个机制还有可
能发生在昆虫的其它部位 ,可能是丝氨酸蛋白酶抑制蛋白调
节酚氧化酶从而使自动防御反应停滞 ,同时有研究表明丝氨
酸蛋白酶抑制蛋白还可以被诱导来抑制酚氧化酶 [46 ] 。不过从
目前国内外的研究情况来看 ,自体免疫通常是一个被研究者
忽略掉的对昆虫免疫防御系统的限制因素。
3. 5 性别
通常在昆虫中雄性的免疫反应能力要低于雌性 [31 ] 。在脊
椎动物中睾酮被视为平衡免疫能力和第二性征表达的媒介 ,
比如 ,雄性为了能更好的吸引雌性就只好牺牲免疫能力 (免疫
活性障碍假说) [47 ] 。昆虫的免疫功能与其第二性征有一定的
联系 ,如蟋蟀鸣叫声音的频率与其体内血细胞数量相关 [48 ] 。
雄性和雌性昆虫在行为学、营养学及被寄生状况都可能
不一样 ,但是造成雌雄免疫能力差别更重要的原因可能是性
别本身。因为雄性所花在繁衍后代上的精力要远远少于雌
性 ,对雄虫而言获得与雌虫交配的机会才是最为重要的。那
么雄虫要加强竞争能力就以降低将来生存能力为代价 ,例如
广泛地和雌性进行交配活动就会削弱雄性祛除寄生物的能
力[49 ] 。另外 ,雌性在被病原感染后所受到的损伤却要高于雄
性[49 ] 。因此 ,雄性在自身的生活史中投入到免疫防御的精力
自然就低于雌性。然而 ,为了种群能够更好地繁衍 ,雌性在选
择配偶的时候就会选择免疫防御能力明显强大的雄性进行交
配[34 ] 。那么诸如雄性蟋蟀的鸣叫声 [48 ]以及豆娘翅的颜色[49 ]
之类的第二性征就为雌性选择配偶时提供了线索来判断配偶
体内是否存在优良的基因。
3. 6 其它因素
昆虫的免疫防御系统还受到其它很多因素的影响 ,只是
对这些因素造成影响的研究还不透彻。比如昆虫免疫防御产
生反应需要一定的时间 ,什么时候开始发生免疫有很大的差
异。当昆虫被感染的几分钟之后酚氧化酶就被激活而且瘤状
物随后出现[50 ] 。同样 ,对于细菌的吞噬作用也是在侵染完成
后的几分钟内就发生 [51 ] 。其它的免疫功能要更多的时间就不
能够阻止病原的定殖和生长。显然 ,鉴于祛除已感染的增殖
速度慢的病原都需要比较长的时间 ,那么昆虫对于增殖速度
很快致病性强的病原就相应的应该有迅速的免疫反应 ,而昆
虫体的免疫防御系统是否与此一致还不清楚 ,环境温度也是
一个影响昆虫免疫防御的重要因素 [52 ] 。另外 ,免疫反应和昆
虫的老化也有一定的联系 ,一个关于蚊子的研究发现蚊子加
速老化可能是其对某种寄生物抗性增强的代价 [53 ] 。最后 ,有
学者认为近亲繁殖可能会导致昆虫免疫能力下降 ,但是这种
说法还缺乏足够的证据。
4 展望
随着分子生物学技术的快速发展 ,昆虫免疫防御机制在
分子水平上的研究已经取得了长足的进步。同时 ,分子技术
的快速发展还使得生态学家和进化生物学家研究昆虫的适应
以及选择的方法上有所革新。另外基因库数据的不断丰富使
得通过在基因水平上比较不同昆虫的免疫防御来验证一些假
设成为可能。另外基因测序和基因表达技术可以查明昆虫免
疫防御和其它功能之间维持平衡的机制及其遗传基础。

自 1962 年 F. Ritossa 发现热休克应答以来 ,有关细胞在高
温、氧化剂、重金属环境和细菌感染等不良条件的应答机制方
面已进行了广泛而深入的研究。Hsps 除了能修复受损伤的蛋
白、降解不能被修复的蛋白和维持新生蛋白的正确折叠外 ,还
有更广泛的细胞生物学功能 ,如在细胞周期 ,胚胎发育 ,细胞
分化 ,精子发生等方面的作用[11 ] 。因此 HSF1 对 Hsps 的调节
作用在细胞中举足轻重。随着对热休克蛋白基因表达的调控
作用机制研究的深入 ,对热休克转录因子的结构功能及调控
HSPs表达中所起的作用将有更多更透彻的了解 ,从而有助于
从分子水平上了解热休克蛋白基因表达的详细机制 ,从而阐
明热休克应答在局部缺血、发炎、感染等多种人类疾病中的启
动机制 ,为寻找和开发能激活细胞热休克防御机制的新型分
子药物提供理论基础 [13 ] 。
