中药蝼蛄提取物对肿瘤细胞的抑制作用初报

摘要:目的:研究蝼蛄提取物对肿瘤细胞的抑制作用。方法:采用结晶紫染色法和MTT法。
结果:以QGY7703细胞作为受试细胞,与阳性对照相比,12个提取物部分中的第7和第11
部分初筛抑制效果达显著水平;运用HL-60细胞对第7和第11部分的抑制效果进一步实验,
二者均具有显著的抑制作用,第7部分大于第11部分。结论:蝼蛄的提取物第7部分具有
显著的抗肿瘤活性。

[Abstract] Objective:To study the inhibition effect of the Louguo(G. orientalis) extract to cancer cell.METH-
ODS: The crystal violet decoration method and MTT method were adopted .RESULTS: The QGY7703 cell was to be experimental cell ,comparing with positive control ,the seventh and the eleventh have marked inhibition
effect to QGY7703 cell. The HL-60 cell was to be experimental cell for further test, the seventh and the eleventh
part have marked inhibition effect to HL-60 cell, the inhibition effect of the seventh part more than that of the
eleventh part. CONCULATION: The seventh part of the Louguo(G. orientalis)extract has remarkable inhibition activity to cancer cell .

[1,5,7]蝼蛄的传统功效是消除各类水肿、利两便、除恶疮、消炎等,近年来在
[6][8,12]临床上有新的药用用途发现,如治疗肝癌后期的腹水、尿潴留,泌尿系统结
[13,14][15]石、消化系统肿瘤等。本实验以QGY7703和HL-60细胞为受试细胞,对蝼
蛄提取物的抑制肿瘤细胞作用进行了初步的研究。

1材料和方法
1.1材料
1.1.1药物和试剂 药物分别用无水乙醇配制成贮液,临用前用RPMI1640培养液稀释。MTT为Serva产品,用PBS溶解成5mg/ml,过滤除菌。10,异丙醇溶液:
(w/v)。 异丙醇用PBS稀释
1.1.1.1结晶紫染色液:结晶紫0.5g,40,甲醛8ml,NaCl 0.17g,乙醇2.3ml,加水至100ml,溶解后混匀备用。
1.1.1.2 结晶紫提取液:柠檬酸三钠0.897g,12N HCl 162.5,l,无水乙醇47.5ml,加水至100ml,溶解后混匀备用。
1.1.1.3 阳性对照:(Sigma公司产品)精确称取7.9888g秋水仙碱用重蒸水溶解,定容至50ml,浓度为0.4mol/L,然后用水稀释为400mmol/L溶液。 1.1.2细胞培养 人白血病HL-60细胞、QGY7703肝癌细胞(联合基因公司提供)。细胞用含有10,新生牛血清(NCS,杭州四季青生物工程材料研究所)的RPMI1640培养液(Sigma公司)传代培养。
1.1.3仪器 DG3022型酶联免疫测定仪(第四军医大学和国营华东电子管厂联营制造)。
1.2方法
1.2.1受试样品:东方蝼蛄和华北蝼蛄的95,的醇提取液、东方蝼蛄95,醇提取液的石油醚萃取部分、醋酸乙酯萃取部分、正丁醇萃取部分、石油醚部分的初分部分以及单体部分等12个部分进行药理学活性筛选。
1.2.2对QGY7703细胞的抑制作用(结晶紫染色法)
4 96孔细胞培养板每孔加入QGY7703细胞(1,10个),在37:C CO孵箱中孵育224 h。加入药物48小时后,结晶紫染色液染色0.5h,洗涤后加入结晶紫提取液100ul,混匀,然后用酶联免疫测定仪测定吸收度值A。 595
1.2.3对HL-60细胞的抑制作用(MTT法)
4 96孔细胞培养板每孔加入100 ,l HL-60细胞(5,10个),然后加入100,l系列稀释的药物,每一药物浓度为三复孔,实验对照为无药培养液(含药物最大 浓度时的相应溶剂量)。细胞在37:C CO孵箱中孵育48 h后,用MTT法测定细胞2
数,即结束培养前4 h加入MTT 10 ,l,然后将细胞培养上清液弃去,加入10 %

异丙醇溶液100 ,l避光放置30 min后,测定吸收度值A值。 570

2结果
实验结果见表1,表2。
选取东方蝼蛄和华北蝼蛄的提取物以及各个萃取部分、初分部分进行药理学
实验,体外QGY7703细胞毒理实验的结果表明,受试药物7和11部分有显著的
细胞毒理效果,其余部分无显著作用。以HL-60细胞为受试细胞,对上述两部分
进一步进行多浓度的重复筛选实验,表2的实验结果表明,7和11部分对HL-60
细胞具有显著的细胞毒作用,7部分的细胞毒性大于11部分。7为11部分的进
一步分离、纯化物,对于第7部分的进一步纯化、鉴定工作正在进行。
